蓝桉超级苗组培快繁技术体系研究

A study on technical system of tissue culture for rapid propagation of Eucalyptus globulus Labill. superior seedlings

  • 摘要: 以蓝桉超级苗茎段为外植体,分别采用正交试验、单因素试验和两因素全因子试验逐一探讨蓝桉外植体消毒、外植体采集时期、腋芽诱导、不定芽增殖和生根培养等组培快繁主要环节中各自的最佳处理,构建蓝桉超级苗组织培养的技术体系. 结果表明:外植体联合消毒的最佳处理为75%酒精消毒20 s,3%洁尔灭消毒1 min和0.05%升汞消毒6.5 min;外植体适宜采集时期为10月份,污染率和褐变率均最低,且存活率和萌芽率均最高;腋芽诱导的最佳体系为改良的MS培养基、0.5 mg/L的KT、0.4 mg/L的6-BA和0.5 mg/L的NAA,可极显著提高腋芽萌发率且诱导效果稳定;不定芽增殖的最佳处理为改良的1/2 MS培养基、0.3 mg/L的6-BA和0.5 mg/L的NAA,可极显著提高增殖系数,使20 d的增殖系数达2.75;生根培养的最佳处理为改良的1/2 MS培养基、0.2 mg/L的NAA,0.5 mg/L的IBA和0.8 mg/L的ABT,可极显著提高生根率和单株生根数量且结果稳定,使生根率达64.38%,单株生根数量达2.70.

     

    Abstract: To explore the best treatment in the main procedures of tissue culture including disinfection, collection period of explants, axillary bud induction, adventitious bud proliferation and rooting culture, and to establish a technical system for tissue culture of Eucalyptus globulus superior seedlings, orthogonal experiment, single-factor experiment and two-factor full factor experiment were used by taking the stem segments of Eucalyptus globulus superior seedlings as explants. The results showed that the optimal treatment for combined disinfection of explants was 75% alcohol disinfection for 20 s, 3% bromo-geramine disinfection for 1 min and 0.05% mercuric chloride disinfection for 6.5 min. The most suitable time to collect explants was in October, during which contamination rate and browning rate were the lowest, survival rate and germination rate were the highest.The best system for axillary bud induction was composed of improving MS medium, 0.5 mg/L KT, 0.4 mg/L 6-BA and 0.5 mg/L NAA, which could significantly increase the germination rate of axillary buds and show stable induction effect. The optimum treatment of adventitious bud proliferation included improving 1/2 MS medium, 0.3 mg/L 6-BA and 0.5 mg/L NAA, which could remarkably increase the proliferation coefficient. The optimal treatment of rooting culture was improving 1/2 MS medium, 0.2 mg/L NAA, 0.5 mg/L IBA and 0.8 mg/L ABT, which could significantly improve rooting rate, increase roots number per plant and the test results were very stable.
