雪茄烟叶霉变真菌蒙地曲霉(Aspergillus montevidensis)的鉴定及其防治研究

Study on the identification and control of Aspergillus montevidensis causing mildew on cigar tobacco leaf

  • 摘要: 鉴定雪茄烟叶霉变真菌种类及筛选安全高效防霉制剂对生产优质雪茄烟叶至关重要. 目前已经报道了引起茄烟叶霉变的多种病原真菌,本研究鉴定云南香料烟有限责任公司甘庄雪茄烟发酵中心的雪茄烟叶原烟及其发酵过程中的霉变真菌并探讨绿色防控技术. 采用可培养方法和柯赫氏法则验证,基于形态学和分子生物学鉴定特征,将雪茄烟叶霉变病原菌鉴定为蒙地曲霉(Aspergillus montevidensis). 蒙地曲霉AM01菌株的最适生长温度为28 ℃,在4 ℃以下或40 ℃以上几乎不生长;AM01菌株生长对氧气浓度需求量不大,在1.5%-15%氧气浓度范围均能生长;在烟叶含水量7%-36.5%范围内,随着含水量增加,AM01菌株引起的霉变病情指数显著增加,烟叶含水量为36.5%时霉变最重. 生物源抑制剂纳他霉素、麝香草酚和抗菌肽对AM01菌株菌丝生长的半抑制浓度(IC50)分别为4.66、15.72 mg/L和42.77 mg/L. 用“1000 mg/L抗菌肽”、“500 mg/L麝香草酚+1000 mg/L抗菌肽”和“1000 mg/L麝香草酚+1000 mg/L抗菌肽”喷雾处理烟叶,发酵28 d时霉变率分别下降了75.55%、77.55%和79.55%;用“1000 mg/L麝香草酚”、“500 mg/L麝香草酚+1000 mg/L抗菌肽”、“1000 mg/L麝香草酚+1000 mg/L纳他霉素”和“1000 mg/L麝香草酚+1000 mg/L抗菌肽”熏蒸处理烟叶,发酵28 d时霉变率分别下降了77.11%、81.12%、79.11%和81.78%.

     

    Abstract: Identification of the pathogen causing mildew on cigar tobacco leaf, and screening of the safe and efficient anti-mildew agents are very important for the production of high-quality cigar tobacco leaves. At present, several pathogenic fungi causing mildew on cigar tobacco leaf have been reported. This study identified the pathogenic fungus causing mildew on cigar tobacco leaf sampling from the Ganzhuang Cigar Fermentation Center of Yunnan Aromatic Tobacco Co., Ltd. and explored the green control technologies. By methods of the culturable and Koch's rule, the mildew pathogen of cigar tobacco leaves sampled from the Ganzhuang Cigar Fermentation Center of Yunnan Aromatic Tobacco Co., Ltd. was identified as Aspergillus montevidensis. The optimum growth temperature for A. montevidensis AM01 was 28 ℃. This strain hardly grew below 4 ℃ or above 40 ℃. Growth of AM01 strain did not require much oxygen, because the growth occurred in the range of 1.5%-15% oxygen concentration. In 7%-36.5% moisture content of the tobacco leaf, the disease index of mildew caused by AM01 strain increased significantly with the increase of moisture content, and the most severe of mildew was recorded at 36.5% moisture content. The IC50 values of natamycin, thymol and antimicrobial peptide on mycelial growth of AM01 strain were 4.66 mg/L, 15.72 mg/L and 42.77 mg/L, respectively. The mildew rates of tobacco leaves sprayed with "1000 mg/L antimicrobial peptide", "500 mg/L thymol+1000 mg/L antimicrobial peptide" or "1000 mg/L thymol+1000 mg/L antimicrobial peptide" decreased respectively by 75.55%, 77.55% or 79.55% after 28 days of fermentation. When fumigated the tobacco leaves with "1000 mg/L thymol", "500 mg/L thymol +1000 mg/L antimicrobial peptide", "1000 mg/L thymol +1000 mg/L natamycin" or "1000 mg/L thymol +1000 mg/L antimicrobial peptide, the mildew rates decreased respectively by 77.11%, 81.12%, 79.11% or 81.78% after 28 days of fermentation.
