EMS mutagenesis in vitro and mutant screening of Echeveria agavoides ‘Corderoyi’

  • In order to determine the optimum treatment method and obtain different types of mutants, Echeveria agavoides ‘Corderoyi’ were treated by ethyl methyl sulfonate (EMS) under different concentrations and soaking time in this study. The pedicels of ‘Corderoyi’ were treated by EMS immersion as explants, and differentiation was induced by tissue culture. Phenotypic difference was observed to screen the mutants. The results showed that the optimum treatment for ‘Corderoyi’ explant mutagenesis was at 0.05% EMS immersion for 6 hours. The fatality rate and mutation rate of explants were 64.86% and 14.50%, respectively. Four types of mutants with stable phenotypic variation and ornamental value were screened. It could enrich their germplasm resources and serve as reference for the mutagenesis of other Crassulaceae plants.
