The establishment of cell suspension culture of rice

  • Immature and mature embryos of 30 rice varieties(lines)were selected and cultured on MS solid medium.According to the callus induction ratio and their response to subculture,they were classified into three groups.CPSLO17 and IR21820-3-2-2-8 showed high callus induction ratio and good response to subculture,were selected to establish the cell suspension culture with N6 liquid medium.The results showed that the genotype is one of the key factors to establish the cell suspensions.With zinc deficiency,the P,Zn contents in suspension cell lines were lower,while P/Zn,Fe/Zn values higher,which is just as the change of mineral elements in rice plant under zinc deficiency.On the other hand,there was little effect on the callus induction frequency,but the differentation ratio decreased under zinc deficiency.
