固相萃取-高效液相色谱测定化感水稻根系分泌的酚酸类化合物

Determination of 10 phenolic acid in root exudates of rice using solid-phase extractionand high performance liquid chromatography

  • 摘要: 建立了固相萃取(SPE)-高效液相色谱(HPLC)测定化感水稻根系分泌的酚酸类化合物的分析方法.选用SPE小柱Cleanert PEP(500mg/6mL)对水稻水培液的酚酸类化合物进行富集.采用Agilent 1260 HPLC色谱仪、ZORBAX SB-C18色谱柱(4.6mm150mm,5m),以甲醇-0.1%磷酸水溶液作为流动相进行梯度洗脱,在流速1.6mL/min、柱温30℃、检测波长280nm的条件下分离测定了3,4二羟基苯甲酸、对羟基苯甲酸、丁香酸、香兰素、香草酸、香豆酸、阿魏酸、苯甲酸、水杨酸、肉桂酸等10种酚酸类化合物.建立的方法成功应用于化感水稻根系分泌的酚酸类化合物的检测,回收率在94.6%~105.8%之间.该方法灵敏度高,重复性好,检测快速、准确,适用于水稻根系分泌的酚酸类化合物的定量测定.

     

    Abstract: This paper developed a method for measuring phenolic acids in root exudates of rice by solid-phase extraction (SPE)-high performance liquid chromatography (HPLC).Phenolic acids were extracted from culture solutions using cleanert PEP (500mg/6mL) column.Chromatographic separation was performed with ZORBAX SB-C18 column(4.6mm150mm,5m)by using the mobile phase gradient elution of methanol-0.1% phosphoric acid mixture,the flow rate is 1.6mL/min,the column temperature is 25℃ and the detection wavelength is 280nm.Under the optimum conditions,10 phenolic acids (3,4-dihydroxybenzoic acid,phydroxy-benzoic acid,vanillic acid,syringic acid,vanillin acid,ferulic acid,coumarin,benzoic acid,salicylic acid,cinnamic acid) were separated and determined.The developed method was successfully used to measure and analyze phenolic acids in the root exudates of rice.The average recoveries were 94.6%105.8%.This method is sensitive,rapid,accurate and with good repeatability.It is suitable for the determination of phenolic acids in the root exudates of rice.
