旺长期烤烟K326内生细菌多样性及部分菌株的酶活筛选

A study on biodiversity and functional enzymes of tobacco K326 endophytes in most productive growth period

  • 摘要: 作者主要研究了旺长期烤烟K326内生细菌的多样性及其产酶特性,共设计8种培养基对不同间作模式下的烤烟样品进行分离,探讨了所分离得到的内生细菌多样性,并对不同培养基的分离效果进行了评价.本研究共分离到193株内生细菌,分布于34个已知属及3个潜在新属.对这些内生细菌分析表明,微生物的多样性较好,其香农指数、辛普森指数及物种丰度分别是2.748,0.25924 和154.9.根据旺长期烤烟K326的生长特征,设计不同的分离培养基,计算多样性指数来判断不同培养基的分离效果,结果表明M5、M6、T3号培养基分离效果最好,M7、Y号培养基次之.按照不同分类单元,从193株分离菌株中挑选出67株,进行酶活力筛选,获得了具有淀粉酶活力的菌株20株(占29.9%)、具有蛋白酶活力菌株19株(占28.4%)、具有木聚酶活力的内生细菌27株(占40.3%)、具有纤维素酶活力的内生细菌29株(占43.3%)、具有甘露聚糖酶活力的菌株12株(占18.0%).结果表明本研究所建立的一套针对烤烟内生细菌的分离方法是行之有效的,分离出大量烤烟内生细菌及具有不同酶活特性的菌株,为后续研究及生产应用奠定了基础.

     

    Abstract: In this research,we explored the diversity of cultured endophytes in tobacco K326 and screening the functional enzymes.Eight kinds of media were designed on different interplanting patterns of tobacco plants,we analysised the biodiversity of the cultured tobacco endophytes and the isolation effect of different medium.193 strains were isolated and these strains were classified into 33 genera and 3 new potential genera.Microbial diversity was relatively rich and the Shannon index,Simpson index,species richness were 2.748,0.25924 and 154.9 respectively.According to the growth characteristics of tobacco K326,we designed eight different media,Shannon index to judge the isolation effect by using different culture medium,the results showed that the isolation effects of M5,M6 and T3 media were good,M7 and Y media were poor.According to different OTUs,we picked up 67 strains from 193 strains for further enzymatic activity screening,and we obtained 20 amylase positive strains(29.9%),19 protease positive strains(28.4%),27 xylanase positive strains(40.3%),29 cellulase positive strains(43.3%)and 12 mannase positive strains(18.0%).The results indicated that the isolation methods which we used were successful for isolating endophytes from tobacco plants,and the new strains we discovered with different functional enzymes would be useful as important resources for further application research.
