不同产地铁皮石斛紫外指纹图谱鉴别分析

Ultraviolet spectrum identification of Dendrobium candidum from different regions

  • 摘要: 采用紫外指纹图谱结合化学计量学方法分析铁皮石斛样品,建立快速鉴别不同产地铁皮石斛的方法.通过单因素实验确定铁皮石斛特征成分提取条件:最适称样量0.2000 g,最佳提取时间30 min;分别用石油醚、氯仿和无水乙醇3种溶剂提取铁皮石斛样品并进行紫外光谱测定,建立不同产地铁皮石斛紫外指纹图谱.结果显示,铁皮石斛的石油醚、氯仿、无水乙醇提取液在24 h内稳定性RSD分别在0~0.99%、0~1.79%、0~0.52%之间,精密度RSD分别在0~0.04%、0~0.04%、0~0.09%,重现性RSD分别在0~0.34%、0~0.30%、0.03%~0.13%,表明该方法稳定、可靠.以样品紫外吸收波长、吸光度值进行偏最小二乘判别分析,结果表明,主成分得分的二维投影图可以准确区分5个产地铁皮石斛样品.该方法能够简单快速地鉴别同一种源不同产地种植的铁皮石斛,为其他物种的产地鉴别提供参考.

     

    Abstract: The ultraviolet fingerprint combined with chemometrics methods were used to analyze the Dendrobium samples and to set up a rapid method for identification of Dendrobium candidum which collected from different origins.It could be used the single factor experiment to determine the extraction conditions of the characteristic components of Dendrobium candidum.The optimal sample weight and the best extraction time were found to be 0.200 0 g and 30 min respectively.Dendrobium samples were extracted with petroleum ether,chloroform and ethanol and the extracting solutions were determined by UV spectra and then UV fingerprint of Dendrobium candidum was set up which were from different origins.The results showed that,Dendrobium candidum extracted by petroleum ether,chloroform and ethanol,in 24 h,the RSDs of stability were 00.99%,01.79%, 00.52%,the RSDs of accuracy were 00.04%,00.04%,00.09% and the RSDs of repeatability were 00.34%,00.30%,0.03%0.13% respectively.It showed that the method was stable and reliable.The analyses of the data have been performed by partial least squares discriminant analysis.The results showed that,the two-dimensional projection of the principal component scores can accurately distinguish Dendrobium samples from 5 areas.This method can be easily and quickly to identify the Dendrobium candidum which were from different regions and belong to the same provenance,and it could provide reference for identification of origins of other species.
