烟叶纤维素降解菌的筛选、鉴定及其产酶条件优化

Screening,identification of cellulose-decomposing strain from aging flue-cured tobacco leaves and optimization of its fermentation condition

  • 摘要: 利用筛选培养基从陈化烟叶中分离到纤维素(CMC)酶活性较高的菌株CE1,对其进行鉴定后表明CE1为绿色木霉 (Trichoderma viride).对其产酶条件进行优化,表明CE1的最佳产酶初始pH为6.5,最适产酶温度28 ℃,培养至第5天时CMC酶活达到最大值.利用其粗酶液对烟丝进行了处理,表明随着处理浓度的提高,烟丝纤维素的降解率也随之升高,烟丝总糖的含量也有所提升.

     

    Abstract: A cellulose-decomposing strain CE1 with high enzyme activity was isolated from aging flue-cured tobacco leaves by selection medium and classified as Trichoderma viride.The optimum pH value of the fermentation of CE1 was 6.5,the optimum temperature was 28 ℃,the carbomethyl cellulase(CMC ase) got the highest activity at the 5th day of culture.Crude enzyme of CE1 was used directly for tobacco processing,the results showed that the degradation rate and total sugar of tobacco increased by increasing the concentration of the enzyme.
