伤痛消胶囊对大鼠长期毒性的研究

An experimental study on long-term toxicity of Shangtongxiao Capsule on rats

  • 摘要: 采用SD大鼠以1.0、2.0、5.0gkg-1 3个剂量灌胃给予伤痛消胶囊内容物26周,停药恢复观察4周,旨在观察其对动物是否产生毒性作用及毒性靶器官,评价长期用药的安全性.结果显示,试验期间各组动物未见异常体征及症状,摄食量、饮水量和体重增长正常;给药13周、26周和恢复观察4周各组雌雄鼠血液学及血清生化学各项指标均无明显异常,主要脏器重量及系数仅呈无规律微波动,相关脏/脑比值组间比较无差异(p 0.05).组织病理学检查仅见给药26周3个剂量组部分雌鼠肾脏近曲小管内皮细胞胞质有少量浅棕色或棕色颗粒沉积,且呈剂量-反应趋势,但停药恢复4周均明显减轻,其它脏器、器官均未见与药物相关的组织形态学异常.未见延迟毒性发生.这些结果提示伤痛消胶囊内容物1.0gkg-1以下为无毒性反应剂量,临床用量的16.67倍以下为安全剂量范围.

     

    Abstract: The aim of this study is to observe the long-term toxicity and possible target organ of Shangtongxiao Capsule contents on rats, also to evaluate its safety of long-term medication.SD rats were randomly divided into three dose groups (1.0,2.0,5.0gkg-1 of Shangtongxiao Capsule contents) and a control group.The rats were consecutively drenched for 26 weeks and then recovered for 4 weeks.No abnormal signs and symptoms were observed in either group.All animals' food intake, water intake and weight gain were normal.There was no obvious abnormality of blood routine and blood diochemistry indicators after administration of 13, 26 weeks or 4 weeks' recovery.The main organs weights and relative coefficient fluctuated slightly.Also,there was no difference in the relative value of viscera/brain between the groups (p 0.05).A little amount of light brown or brown particles deposition in renal proximal tubule endothelial cytoplasm was dose-related observed in a few female rats by histopathological examination after 26 weeks' administration,which were reduced after 4 weeks recovery.There was no obvious morphological abnormality in other drug-associated visceras and organs.No delayed toxicity was observed.The results indicated that 1.0gkg-1 of Shangtongxiao Capsule content is NOEAL(no observed adverse effect level).The safety dose range is 16.67 times below the clinical dosage.
