佛甲草细胞毒活性成分研究

  • 摘要: 采用半制备高效液相和中压柱色谱方法对佛甲草的化学成分进行纯化分离,并利用现代波谱技术鉴定单体化合物的结构,并采用MTT法对所得到的化合物进行细胞毒活性筛选.从佛甲草甲醇提取物中分离到9个纯化合物,分别鉴定为山奈酚(1)、木犀草素(2)、3,5,7,3',5'-五羟基黄酮(3)、槲皮素(4)、紫云英苷(5)、咖啡酸(6)、阿魏酸(7)、4-羟基苯甲酸(8)和β-谷甾醇(9).化合物1~8均为首次从该属植物中分得.体外细胞毒活性评价结果显示化合物1~7对HepG2和Hep3B两个肝癌细胞株有不同程度的抑制作用.其中,苯丙酸类成分6~7比黄酮类成分1~5显示出强的细胞毒活性,IC50值范围为19.7~56.4μg/mL.

     

    Abstract: To discover new cytotoxic constituents from the traditional Chinese folk medicine “Fo Jia Cao” (whole plants of Sedum lineare Thunb.),the isolation and purification of a methanolic extract was performed with medium-pressure column chromatography and preparative HPLC.Chemical Structures of isolated compounds were elucidated by detailed spectral analysis.Cytotoxic activity of compounds was evaluated using the MTT method.Nine pure compounds were obtained and their structures were elucidated as kaempferol (1),luteolin (2), 3,5,7,3',5'-pentahydroxyflavone (3),quercetin (4),astragalin (5),caffeic acid (6),ferulic acid (7),4-hydroxybenzoic acid (8) and β-sitosterol (9).Compounds 1—8 were firstly isolated from the title plant. Compounds 1—7 displayed cytotoxic activity.Phenylpropanoid acids 6 and 7 exhibited better cytotoxic activity than flavonoids 1—5 with IC50 values ranging from 19.7 to 56.4μg/mL.
