不同pH、温度和储藏因素对云南松针叶碳酸酐酶稳定性的影响

Research on the stability of pH values,temperatures and storage factors of carbonic anhydrase in Pinus yunnanensis needles

  • 摘要: 试验以5种不同pH缓冲液处理的4个不同年龄云南松针叶中的碳酸酐酶为研究对象,分析不同pH环境对云南松碳酸酐酶稳定性的影响;以4个不同年龄云南松针叶中的碳酸酐酶为研究对象,分析其在6种不同温度处理下的活性变化,探究云南松碳酸酐酶的热稳定性;对4个不同年龄云南松的针叶进行储藏,分析储藏温度和储藏时间对云南松针叶碳酸酐酶储藏稳定性的影响.结果表明:① 不同年龄的云南松针叶碳酸酐酶皆在碱性条件下能够保持较好的稳定性;② 在常温下,云南松针叶碳酸酐酶具有良好的热稳定性;③不同龄的云南松针叶碳酸酐酶相对活性随储藏温度和储藏时间的变化规律基本相同,4年、8年和14年生,处理组合5的相对酶活性皆最大,4年、8年和22年生,处理组合16的相对酶活性皆最小;4年、8年和14年生云南松,影响其针叶碳酸酐酶活性变化的主要因子是储藏时间,22年生云南松,影响其针叶碳酸酐酶活性变化的主要因子是储藏温度.

     

    Abstract: The stability of carbonic anhydrase of Pinus yunnanensis needles in different pH environment was analyzed through the test which the carbonic anhydrase in the needles of P.yunnanensis at four different ages were processed by buffers at five different pH.The results showed that carbonic anhydrase were able to maintain good stability in alkaline condition.At the same time,the stability of carbonic anhydrase of P.yunnanensis needles at different temperatures was analyzed and the results showed that carbonic anhydrase had good thermal stability at room temperature.The Orthogonal test design was adopted to research the storage stability of carbonic anhydrase.The results showed that the rules of relative activity of carbonic anhydrase changing by different storage temperatures and times were basically the same in four different ages.Among 4a,8a and 14a,the relative activity of carbonic anhydrase was the highest by the treatment combinations NO.5.Among 4a, 8a and 22a,the relative activity of carbonic anhydrase was the lowest by treatment combinations NO.16.For 4a,8a and 14a,the main factor which affected the carbonic anhydrase activity was the storage time,and for 22a,the main factor was the storage temperature.
