紫外光谱法研究鬼臼毒素与环糊精及其衍生物的包合行为

Studies on the inclusion behavior of podophyllotoxin with cyclodextrin and its derivatives by ultraviolet spectroscopy

  • 摘要: 采用紫外—可见光谱滴定法分别测定了主体分子环糊精及其衍生物与客体分子鬼臼毒素在室温,pH=3.0和pH=10.5的缓冲溶液中形成配位包合物时的稳定常数(Ks).结果表明:鬼臼毒素与环糊精(α-CD,β-CD,γ-CD,DM-β-CD和TM-β-CD)形成包合时,碱性条件下稳定常数(Ks)相对更大,鬼臼毒素在碱性条件下更稳定.主体-客体之间的尺寸匹配在包合形成的过程中起到决定性的作用,γ-CD空腔尺寸大小与客体分子鬼臼毒素的尺寸匹配程度较好,故其Ks值最大.此外,β-CD环糊精的取代基也影响主体配位能力,鬼臼毒素与甲基化β-CD形成包合时,环糊精上甲氧基取代越多,越不利于包合的形成.

     

    Abstract: Spectrophotometric titrations were performed in aqueous buffer solution (pH=3.0,pH=10.5) at 25℃ to determine the binding constants of cyclodextrins and its derivatives with podophyllotoxin guest molecules.The results indicate that podophyllotoxin with cyclodextrins and its derivatives in alkaline conditions (pH=10.5),the binding constant are relatively large.Podophyllotoxin is more stable in alkaline conditions under the formation of inclusion complexes;the molecular inclusion ability is dominated by the size/shape-matching between host and guest,the size/shape of γ-CD cavity and guest molecules podophyllotoxin are well matched,so the Ks is maximum.In addition,the substituent on the β-CD derivatives changes its original binding ability,podophyllotoxin with methylated β-CD form the inclusion complex,the methoxy is substituted more on β-CD,less well to the inclusion formation.
