基于纳米多孔铂检测细胞中多巴胺

  • 摘要: 通过还原反应合成纳米多孔Pt材料,并对其进行表征.纳米多孔Pt作为一种纳米模拟酶,对过氧化氢氧化3,3',5,5'-四甲基联苯胺(TMB)形成蓝色氧化产物的反应具有良好的催化作用.加入多巴胺(DA)后,由于DA吸附到纳米多孔Pt的孔道上,减少了催化位点,对纳米多孔Pt的催化作用产生抑制效应,使形成的TMB蓝色氧化产物减少,导致吸光度值下降.研究利用多巴胺对纳米多孔Pt催化作用的抑制效应,采用分光光度计检测体系的吸光度从而实现多巴胺的定量检测.考察了过氧化氢浓度,多孔Pt浓度以及多巴胺反应时间对吸光度的影响.在最优条件下,吸光度值与多巴胺浓度有良好的线性关系,线性范围为0.08~1.0mmol/L,相关系数为0.9959,检测下限为0.0085mmol/L.该体系为多巴胺的检测提供了一种简单快速的方法,并被用于黑色素瘤细胞裂解液样品中多巴胺的检测.

     

    Abstract: In this paper,nano-porous Pt materials was synthesized by reduction reaction,and was characterized by TEM.Dopamine can inhibit the catalytic activity of bio-enzyme.Nano-porous Pt materials,as a nano-mimic enzyme,was inhibited by dopamin as well.Porous Pt NPs have a good catalytic activity on the reduction of H2O2,and the above system combined with TMB will have a chromogenic reaction.In this study,inhibitory effect of dopamine to the catalysis of porous Pt NPs was used to realize the quantitative detection of dopamine using ultraviolet spectrophotometer (UV-Vis) to detect the absorbance of the system.The concentration of H2O2,porous Pt NPs and the reacting time between porous Pt NPs and DA were optimized.Under the optimal experimental conditions,the detection range of the system is from 0.08 mmol/L to 1.0 mmol/L with a good linear correlation coefficient of 0.9959 and the detection limit of 0.0085 mmol/L.This system provides a new,simple as well rapid method on the detection of DA and can be used for the detection of DA in cell.
