解淀粉芽孢杆菌的GFP标记及土壤抑细菌作用分析

GFP tagged Bacillus amyloliquefaciens and analysis on the soil bacteriostasis

  • 摘要: 土壤抑细菌作用是影响生防细菌定殖的关键因素之一.土壤抑细菌作用检测方法的建立对开拓土壤抑细菌作用领域的研究极为重要.作者以生防菌解淀粉芽孢杆菌ZY-9-13为材料,通过GFP蛋白标记,建立了通过双抗平板及菌落荧光计数定量检测土壤抑细菌作用的方法.测定了云南省10个州市30份烟田土壤对Bacillus amyloliquefaciens ZY-9-13-gfp的抑菌作用.结果表明供测土壤的平均抑制率为23.92%.供测土样的pH值、有机质含量、微生物多样性指数与抑菌率呈正相关关系,相关系数r分别为0.79、0.90和0.93.

     

    Abstract: Soil bacteriostasis is one of the important factors which influence the colonization of biocontrol bacteria in soil.A method of measuring the soil bacteriostasis is vital for further studies in this area.In this study,B. amyloliquefaciens ZY-9-13 was tagged by green fluorescent protein gene (gfp),and the soil bacteriostasis of 30 tobacco soil samples collected from 10 cities of Yunnan Province was measured based on dual-resistance medium and fluorescent colony.The results showed an average value of 23.92% of soil bacteriostasis.pH values,organic matters and microorganism diversity index of tobacco soil showed positive correlation with soil bacteriostasis,with coefficient values of 0.79,0.90 and 0.93,respectively.
