树莓叶片中主要抗氧化物质提取工艺优化研究

Study on optimization of extraction technology of main antioxidant substances in raspberry leaves

  • 摘要: 以澳洲红树莓叶片为研究材料,利用超声辅助乙醇提取法同时考察总黄酮含量、超氧化物歧化酶(Superoxide Dismutase,SOD)总活力和总蛋白含量3个指标.基于单因素实验结果,对乙醇质量分数、料液比、超声时间采用Box-Behnken 实验设计和响应面法(RSM)优化提取条件,从而获得最佳的混合提取工艺.再用此工艺比较不同品种树莓间叶片中黄酮类化合物、SOD总活性和总蛋白含量的高低.实验获得的最佳提取工艺参数为:乙醇质量分数60%、料液比1∶40g/mL,超声时间30min,验证提取得到:产物总黄酮含量51.74mg/g、SOD总活力为5828.96 U、总蛋白质量分数为17.20mg/g,实验结果与模型预测值接近,表明最佳工艺条件可行.同时将最佳提取工艺用于黑红莓、托拉蜜、诺娃(有刺)、诺娃、海尔特兹、来味里、马拉哈丁以及秋来斯和澳洲红共9个树莓品种,结果显示黑红莓、诺娃(有刺)和诺娃的叶片总黄酮含量、SOD总活力和总蛋白含量较高,海尔特兹和来味里较低,这与总抗氧化活性测定的结果相符.这些结果为树莓叶片醇提物的研究和引种选育工作提供数据支持.

     

    Abstract: In this study,firstly,the leaves of Australian raspberry were used as materials,and the contents of total flavonoids,total SOD activity and total protein of leaves were investigated by the ultrasonic assisted ethanol extraction process.Based on the single factor experimental results of ethanol concentration,liquid ratio and ultrasonic time,Box Behnken experimental design and response surface methodology(RSM)were selected to further analyze and optimize the mixedextraction conditions.The finnal extraction process was 60﹪ethanol,solid-liquid ratio 1∶40(g / mL)and 30 minutes ultrasound. The optimum values were the total flavonoid content of 51.74 mg/g,SOD total activity of 5828.96 U and total protein content of 17.20 mg/g.Secondly,the optimum process was applied to 9 raspberry cultivars,including Torremi,Nova(with thorns),Nova(without thorns ),Heritage,Reveille,Mara Harding and Autumn Bliss(from the experimental field of Beijing Agricultural College,Beijing)and Australian raspberry(from the experimental field of North University of China,Shanxi),and the results showed that total flavonoids,total SOD activity and total protein contents of Black-red raspberry,Nova(with thorns)and Nova were higher than those of others,and those of Reveille and Heritage were almost the lowest.Similarly,total antioxidant activities of black raspberry and Nova(with thorns)were higher than those of Heritage and Reveille by DPPH analysis.Overall the results provided support for the introduction,selection and comprehensive utilization of raspberry.
