芦笋老茎皂苷对小鼠实验性高脂血症的预防作用*

The preventive effect of the saponins from Asparagus officinalis L. by-products on experimental hyperlipidemiain mice

  • 摘要: 探究了芦笋老茎皂苷(saponins from Asparagus officinalis L.by-products,SAO)对小鼠实验性高脂血症的预防作用.小鼠先饲喂基础饲料同时预防性地给予SAO 14d,15d时再改以高脂饲料诱发高脂血症并继续灌胃相应剂量SAO 42d.结果显示,和高脂对照组相比,预防性给予40,80mg/kg和160 mg/kg的SAO都能明显降低小鼠血清总胆固醇和低密度脂蛋白胆固醇,但不存在明显的剂量依赖关系.SAO还能显著提高小鼠肝脏肝酯酶活性,提示通过提高肝酯酶活性来加强胆固醇清除可能是SAO调节脂代谢的途径之一;此外,SAO还能提高小鼠肝脏超氧化物歧化酶和总抗氧化能力,降低血清天冬氨酸氨基转移酶和丙氨酸氨基转移酶水平,说明SAO可能通过提高机体的抗氧化能力,缓解氧自由基对肝脏的损伤,改善肝脏的功能,从而加快脂质的代谢和清除.

     

    Abstract: Asparagus (Asparagus officinalis L. ) by-products,i.e.the parts of the spears discarded during industrial processing,might have potential as food supplements for the therapeutic effects.In this study,the preventive effect of the saponins from Asparagus officinalis L. by-products (SAO) on experimental hyperlipidemia were examined by using mice model.Mice were fed a standard chow diet and treated with SAO at three different doses (40,80 or 160 mg/kg body weight) for 14 days.Then hyperlipidemia in mice was induced by high-fat diet and SAO was administered to mice at the same dose level for 42 days.The results showed that SAO markedly reduced the level ofserumtotal cholesterol and serum low density lipoprotein cholesterol.Also,SAO increased the hepaticlipase activity of liver cells,which indicated that SAO can raisehepaticlipase activity which increase the conversion of cholesterol to bile acids in liver.Finally,SAO decreased the activity of alanine transaminase and aspartate transaminase in serum and increased superoxide dismutaseactivity and total antioxidation capacity in liver which implied that SAO has strong hypolipidaemic and hepatoprotective effect by restoring the antioxidant defense system and ameliorating the liver damage.It issuggested that SAO has strongpreventive effect on experimental hyperlipidemia miceand could be considered as a supplement in healthcare foods and drugs for human.
