10种报春花亲缘关系的ISSR分析

ISSR analysis on the inter-species relationship in Primulas

  • 摘要: 利用简单重复序列间(ISSR)标记对10种报春花的种间变异进行了研究,从40个引物中筛选出11个能扩增出清晰带型并具多态性的引物,共扩增出了86条DNA片段,分子量在300~2000bp之间,平均每条引物扩增出7.81条DNA片段.采用POPGENE软件,计算了种间的Nei氏距离,并用UPGMA法构建了系统树,结果表明:这10种报春花明显聚为三大类:即鄂报春,无粉头序报春,灰岩报春聚为一类;滇北球花报春,报春花、欧报春聚为一类;香海仙报春,橘红灯台报春,霞红灯台报春,海仙花聚为一类.其中第3类与形态学分类结果一致.并探讨了香海仙报春与海仙花的关系.

     

    Abstract: Inter-species relationship of 10 species in Primula were studied by using of ISSR marker.Eleven primers were screened out from forty random primers,and a total of 86 DNA fragments were amplified ranging from 300-2000bp.The average number of DNA fragments produced by each primer was 7.28.Nei’similarity coefficients and genetic distances were caculated by using POPGENE software and the dendrogram were constructed by using UPGMA.The 10 Primula species were divided into three groups.P.poissonii Franch,P.Wilsoni Dunn,P.bulleyana Forr.,P.beesiana Forr.val.beesiana were clustered into one group,which is in consistence with the morphological classification.The relationships of P.poissonii Franch.and P.wilsoni Dunn.were discussed.
