放线菌生物活性物质的高通量筛选

High throuphput screening of bioactive substances from actinomycetes

  • 摘要: 放线菌是产生生物活性物质最多的一类微生物.最近3年,我室从云南省采集土样,分离到3000多株放线菌、真菌,选择其中2600株,进行发酵,制备成提取物,利用自建的抗氧化、抑制乙酰胆碱酶、抑制一氧化氮合成酶、AGE等模型和高通量筛选系统进行了筛选,筛选到一些值得进一步研究的菌株.对高通量筛选的有关问题及初步筛选结果进行了讨论.

     

    Abstract: Actinomycetes are one of important microbial resources producing bioactive metabolites.2600 actinomycete strains were isolated from soil samples collected from Yunnan.Each strain was fermented and extracted,and 2600 extracts were obtained.These extracts were selected by using 4 models in vitro with High Through Screening system.Results of screening indicate that 0.4% strains have anti-oxidation,and 2.7%,(0.34%) and 0.3% of strains have inhibitions against AchE,NOS,and AGE respectively.Inhibition of strain YIM 32766 against AchE was 108.3%.Inhibition of one of elements of extracts of strain YIM 30243 against NOS was 151.8%.Inhibition of strain YIM32012,YIM32215,YIM32234,YIM32212,YIM32044 and YIM32025 against AGE were 300.66%,42.16%,133.94%,122.59%,110% and 101.71% respectively.These strains with high bioactivity should have potential value for future development of drug.
