Abstract:
A high performance liquid chromatography(HPLC) method for the determination of sugar,glycerol and ethanol in ratafee was studied.Ratafee was preseparated by solid phase extraction with Sep Pak C 18 column.Sugars,glycerol and ethanol were separated on a Waters Sugar Pak Ⅰ(6.5mm×300mm) cations ion exchange column,with a refractive index detector system and 0.05g/L EDTA calcium disodium salt solution as mobile phase.Sucrose,glucose,fructose,glycerol and ethanol could be simultaneously determined in a single injection.The limits of detection were:sucrose 1.5μg/mL,glucose 2.0μg/mL,fructose 2.0μg/mL,glycerol 2.5μg/mL and ethanol 2.5μg/mL.The mean recoveries of sugar,glycerol and ethanol were between 97% and 105% and the relative standard deviations were between 0.92%and 1.52%.This method can be applied to the determination of sugars,glycerol and ethanol in ratafee with satisfactory result.
