反义ACC合成酶基因导入番茄的研究

  • 摘要: 采用农杆菌介导法将携带有反义ACC合成酶基因和NPT II的表达载体pKAC导入番茄栽培品种美国大红五星102、美国大红108、美国大红王、大红II号等中.经过卡那霉素筛选后,获得了一批抗性苗,其中有3株PCR检测为阳性.实验证明,不同激素配比对番茄子叶不定芽的诱导有一定影响,适宜的农杆菌侵染时间和浓度是番茄基因转化成功与否的关键,延迟选择有利于番茄基因转化细胞的生长.

     

    Abstract: An expressing vector pKAC with antisense ACCase gene and NPT II was transferred into some tomato strains by agrobacterium mediated transformation.After selection culture with kanamycin,some transgenic plants of tomato were obtained.Three of them were positive by PCR analysis.The results showed three characteristics,namely,1.The inductive rate of adventitious bud of tomato cotyledon was different on various media with different hormone and concentration.2.The concentration of agrobacterium and time of agrobacterium infection were the keys to gene transformation of tomato.3.The delayed selection culture was helpful to the growth of transgenic cells.
