一株产纤维素酶温泉高温菌的分离鉴定及发酵条件优化

Isolation and identification of a cellulose degrading thermophile bacterium andits fermentation cellulase characterization

  • 摘要: 为获得高产高温纤维素酶菌株,从云南元江瓦纳温泉中筛选分离得到一株产纤维素酶的高温菌ZZS-12.该菌在固体培养基上形成白色半透明圆形菌落,边缘圆滑,表面黏稠,经生理生化及16S rRNA序列分析,鉴定为Paenibacillus属的一个新种细菌.该菌最佳碳源、氮源及无机盐分别为麸皮、NaNO3及NaCl.最佳产酶培养时间为60h,最适温度为45℃,最适pH为7.

     

    Abstract: A cellulose degrading thermophile bacterium ZZS-12 was isolated from Wana hot spring in Yunnan province.ZZS-12 on the solid medium A formed a white translucent circular colonies with the smooth edges and the sticky surface.Its physiological and biochemical properties were investigated,and its 16S rDNA sequences were identified,the bacterium was identified as Paenibacillus sp.nov..Best enzyme production of the bacteria fermentation time is 60h.The best carbon,nitrogen sources and mineral salts were bran,NaNO3 and NaCl.The optimum temperature was 45℃,the optimum fermentation time was 60h and the optimum pH was 7
