一株产纤维素酶温泉高温菌的分离鉴定及发酵条件优化

摘要: 为获得高产高温纤维素酶菌株,从云南元江瓦纳温泉中筛选分离得到一株产纤维素酶的高温菌ZZS-12.该菌在固体培养基上形成白色半透明圆形菌落,边缘圆滑,表面黏稠,经生理生化及16S rRNA序列分析,鉴定为Paenibacillus属的一个新种细菌.该菌最佳碳源、氮源及无机盐分别为麸皮、NaNO3及NaCl.最佳产酶培养时间为60h,最适温度为45℃,最适pH为7.Abstract: A cellulose degrading thermophile bacterium ZZS-12 was isolated from Wana hot spring in Yunnan province.ZZS-12 on the solid medium A formed a white translucent circular colonies with the smooth edges and the sticky surface.Its physiological and biochemical properties were investigated,and its 16S rDNA sequences were identified,the bacterium was identified as Paenibacillus sp.nov..Best enzyme production of the bacteria fermentation time is 60h.The best carbon,nitrogen sources and mineral salts were bran,NaNO3 and NaCl.The optimum temperature was 45℃,the optimum fermentation time was 60h and the optimum pH was 7
