一种简便的STR重复次数测定方法

A kind of convenience STR unit measure method

  • 摘要: 短串联重复(STR)的多态性分析是生物个体识别最有效的一种手段,在法医学鉴定中已被广泛用于身份识别.但目前使用的方法成本太高,以致不便在更广泛的范围推广.针对法医上常选用的4 bp重复单位的STR基因座,采用PCR产物小于160 bp的迷你STR(Mini STR)和优化聚丙烯酰胺凝胶电泳条件,提高分辨率,使重复单位之间能有效分辨.设计PCR引物,使STR基因座扩增产物的大小为4 bp的倍数,应用8 bp的DNALadder即能精确测定整数倍STR重复单位数.这一方法操作简便,成本低廉,尤其是用于制作人基因身份证,使得人人制作一张基因身份证成为可能.

     

    Abstract: The polymorphism of Short Tandem Repeat(STR) have been used in individual identifications,especially in medicolegal investigations.But such technologies are too expensive to be preformed widely.The 4 bp repeats unit STR site which was used commonly in medicolegal investigation were chosen to design the new PCR system to limit the PCR products length as 4 bp multiple and less then 160 bp,a Mini STR technique,and optimized the PAGE conditions to distinguish the single STR repeat unit accurately.Then the number of STR repeat units can be measured the numbers of the integer multiple alleles accurately and easily by using 8bp DNA ladder.The new methods preformed in analyze of polymorphism of STR,a simpler and cheaper methods,is suitable to make human gene identification card(ID-card) and it is possible that each people hold a gene ID-card.
