锌试剂-蛋白质体系的共振光散射光谱研究

A study on interaction of zincon with proteins by resonance light scattering measurements

  • 摘要: 在pH 3.0的B-R缓冲溶液中,锌试剂能与蛋白质结合形成复合物.此结合反应能显著加强锌试剂的瑞利光散射信号.详细研究了此结合反应的最佳反应条件,并以此反应为基础,利用共振瑞利散射光技术,建立了一个测定蛋白质的新方法.该方法对牛血清白蛋白(BSA)、人血清白蛋白(HSA)以及免疫球蛋白测定的线性范围分别为0.25~12.5,0.10~15.0μg/mL和0.10~12.5μg/mL,检出限均小于0.05μg/mL,且大量的常见金属离子、氨基酸等共存物质不干扰测定.方法具有很高的灵敏度、很好的选择性及重现性.用于血清样品中蛋白质的测定,结果满意.

     

    Abstract: In B-R buffer solution of pH 3.0,zincon can bind to the proteins form a complex,which results in a remarkably enhancement of the Rayleigh light-scattering(RRLS) signal of zincon.The optimum reaction conditions has been studied in detail,and a new method for the determination of proteins by the resonance Rayleigh light-scattering technique has been developed based on this interaction.The linear range of the method for the determination of bovine serum albumin(BSA),human serum albumin(HSA) and Immunoglobulin G(IgG) are 0.25—12.5 μg/mL,0.10—15.0 μg/mL and 0.10—12.5 μg/mL with a detection limit all below 0.05 μg/mL,respectively.Moreover,a lot of amino acids and metal ions do not interfere in the determination.The method with high sensitivity,good selectivity and reproducibility has been satisfactorily applied to the determination of the total protein in human serum samples.
