Abstract:
The internal transcribed spacer(ITS) regions of ribosomal DNA of 8 Amanita species from Tibetan and Yunnan in China were sequenced by PCR amplication,and their sequences were analyzed by the software of Clustal X(version 1.81).The results showed that,the ITS sequences of 8 species had significant differences in both length and bases,besides the fragment of 5.8S rDNA,whose length is 160bp,being relatively conservative;the length of these fragments of ITS1,ITS2,were 206—297bp,191—238bp,respectively.These implied that the genetic diversity among the Amanita species was relatively high.The 8 Amanita species was dividied into 3 groups at D=0.14 according to the phylogenitic analysis by MEGA and it was dissimilar to traditional taxonomy.The results above provided a molecular reference for further taxonomy and study of Amanita in the future.
