固相萃取和高效液相色谱法测定鱼腥草中的黄酮

Study on determination of flavonoid in houttuynia cordata thunb by solid phase extraction and RP-HPLC

  • 摘要: 研究了用固相萃取预分离,高效液相色谱法测定鱼腥草根和鱼腥草叶中黄酮类物质的方法.鱼腥草根和鱼腥草叶中的黄酮用80%乙醇溶液加热回流提取,提取液用Waters Sep-Pak-C18固相萃取小柱预分离脱脂,以Waters Nova-Pak-C18(Φ3.9 mm×150 mm,5μm)色谱柱为固定相,以0.05 mol/L磷酸二氢钾缓冲溶液和甲醇的体积比为40:60的混合液为流动相,在该色谱条件下,鱼腥草中主要的黄酮成分均达到基线分离;用紫外二极管矩阵检测器在360 nm波长处检测,并作了色谱峰纯度分辨.方法标准回收率为98%~101%,相对标准偏差为0.85%~2.2%.

     

    Abstract: A high performance liquid chromatography(HPLC) method for the determination of flavonoid in myrica rubra leaf and bark was studied.The flavonoid was extracted from the sample with 80% of ethanol by heating circumfluence.Then the extract was degreased by solid phase extraction with Sep-Pak-C18 cartridge.The flavonoid was separated on a Nova-Pak-C18 chromatogram column(3.9×150mm,5μm),with methanol and 0.05mol/L potassium dihydrogen phosphate buffer solution as mobile phase at 60:40(V/V),and monitored with the photodiode array detector at 360nm.The recoveries of the flavonoid are 98%-101%,and relative standard deviations are 0.85%-2.2%.
