抗盐酸克伦特罗单克隆抗体的制备和鉴定

Preparation and identification of monoclonal antibody against β-adrenergic agonist clenbuterol hydrochloride

  • 摘要: 以重氮反应将盐酸克伦特罗与牛血清白蛋白、鸡卵清蛋白偶联制备免疫原和检测原,通过杂交瘤技术获得了3株能分泌特异结合CL小分子的抗体杂交瘤细胞株.经竞争法ELISA分析,9E3抗体的IC50质量浓度为19.85μg/L,14C5抗体为1.65μg/L,16F4抗体为0.84μg/L.在与类似物的交叉性反应中,16F4抗体与沙丁胺醇的交叉反应性为0.26%,特布他林为0.04%,与非诺特罗、肾上腺素、去甲肾上腺素的交叉反应性都小于0.02%.因此,所制备的抗盐酸克伦特罗的抗体的特异性高,可用于与其相关的免疫检测研究和应用.

     

    Abstract: Three hybridomas secreting antibodies against specifically clenbuterol hydrochloride(CL),(β-adrenergic)agonist,which was diazotised and coupled to bovine serum albumin(BSA)to prepare immunogen,and to ovalbumin(OVA)to prepare coating conjugates,were produced by using hybridoma technology.By using competitive ELISA,the 50% inhibition concentration of free CL for 9E3,14C5 and 16F4 antibodies were 19.85,1.65?μg/L and 0.84?μg/L,respectively.The 16F4 antibody has approximately 0.26% cross reactivity to salbutamol,and approximately 0.04% to terbutalin,and no cross reactivity(0.02%)to fenoterol,adrenalin and noradrenalin.Therefore,these antibodies have high specificity for CL and could be used in immunological research and application.
