UPLC-MS/MS法同时测定云南黄草乌中滇乌碱、粗茎乌头碱甲和塔拉萨敏的含量

Simultaneous determination of the contents of yunaconitine, crassicauline A and talatisamine in Aconitum vilmorinianum Kom from Yunnan by UPLC-MS/MS method

  • 摘要: 用UPLC-MS/MS法同时测定云南5种不同产地黄草乌中滇乌碱、粗茎乌头碱甲和塔拉萨敏的含量.采用安捷伦C18色谱柱,以乙腈-甲酸(φ=0.1%)水溶液为流动相进行梯度洗脱,电喷雾离子源正离子多反应监测模式进行质谱分析,采用多个特征离子对和外标标准曲线法对3种生物碱进行定性、定量分析.滇乌碱、粗茎乌头碱甲和塔拉萨敏在质量浓度1~100ng·mL-1范围内线性良好(r≥0.9990),检出限(LOD)分别为0.01、0.01ng·mL-1和0.02ng·mL-1,定量限分别为0.03、0.04ng·mL-1和0.07ng·mL-1;准确度均大于90%,日内及日间精密度均小于5%.5种黄草乌中均检出滇乌碱、粗茎乌头碱甲和塔拉萨敏,但不同产地黄草乌中3种成分的含量差异较大.

     

    Abstract: To establish an UPLC-MS/MS method for simultaneous determination the contents of yunaconitine,crassicauline A and talatisamine in the five roots of Yunnan Aconitum vilmorinianum Kom,the analytical method has been developed on Agilent C18 column and the mobile phase containing ACN and 0.1% FA in water which was running in gradient elution way.The mass spectrometric acquisition was carried out by electospray ionization in positive mode with multiple reaction monitoring method,multiple characteristic ion pairs and the external standard method were opted to qualitative and quantitative analysis for yunaconitine,crassicauline A and talatisamine respectively.The linear ranges were 1—100ng·mL-1 for yunaconitine,crassicauline A and talatisamine (correlation coefficient were over 0.9990).The detection limits were 0.01,0.01ng·mL-1 and 0.02ng·mL-1 respectively;The quantitation limits were 0.03,0.04ng·mL-1 and 0.07ng·mL-1.The accuracies were all higher than 90%,The intra- and inter-day precisions were all less than 5%.Yunaconitine,crassicauline A and talatisamine were all detected in five roots of Yunnan Aconitum vilmorinianum Kom.It all contains yunaconitine,crassicauline A and talatisamine in five different roots of Yunnan kusnezoff monkshood,but the different roots contain different amount of yunaconitine,crassicauline A and talatisamine.
