Study on the synthesis of o-veratraldehyde

  • 摘要: 邻藜芦醛是一类重要的医药中间体,在总结其常见合成方法的基础上,提出了以愈创木酚为原料,经酚羟基烯丙基化、克莱森重排、酚羟基甲醚化、烯烃异构化反应和臭氧氧化/还原反应合成邻藜芦醛的方法,5步反应的总产率为54%.此合成路线原料价廉易得,反应可操作性强,后处理简单,有一定的工业应用价值.

     

    Abstract: o-Veratraldehyde is an important pharmaceutical intermediate.With common synthetic methods taken into consideration,veratraldehyde was prepared by using guaiacol as the starting material and conducting a series of phenolic hydroxyl allylation,Claisen rearrangement,phenolic hydroxy methyl etherification,Triton B-catalyzed olefin isomerization and ozonation/reduction reactions.The total yield of this 5-step reactions was 54%.The above-mentioned synthesis method only requires raw materials that are low-cost and easily acquired,its relative reaction conditions are mild and the work-up is simple,thus it may provide certain value in the industrial applications.
