Effect of the 5′untranslated region sequence of the HSV-1immediate early gene as the 5′UTR of the model protein mRNA on mRNA translation

  • 摘要: 为了探究HSV-1即刻早期基因的5′非翻译区序列作为报告基因EGFP mRNA的5′UTR对其翻译的影响,本研究通过细胞和动物报告基因EGFP的表达水平检测,分析比较四个即刻早期基因5′UTR以及β珠蛋白5′UTR作为mRNA的5′UTR的表达效率. 经动物实验和细胞实验发现:RL2的5′非翻译区序列作为报告基因EGFP的5′UTR的表达水平与β珠蛋白5′UTR相近,RS1、US1、US12的5′非翻译区序列作为报告基因EGFP的5′UTR,表达水平明显弱于β珠蛋白5′UTR. RL2的5′非翻译区序列有潜力作为mRNA疫苗的5′UTR.

     

    Abstract: In order to explore the effect of the 5 'untranslated region sequence of HSV-1 immediate early gene as the 5' UTR of model protein mRNA on its translation, this study analyzed and compared the expression efficiency of 4 immediate early genes 5 'UTR and βglobin 5' UTR as the 5 'UTR of mRNA by detecting the expression level of EGFP in cells and animals. The results of animal and cell experiments showed that the expression level of 5 'UTR of RL2 as model protein was similar to that of β-globin, and the expression level of 5' UTR of RS1, US1, US12 as model protein was significantly lower than that of β-globin 5 'UTR. The 5 'untranslated region sequence of RL2 has the potential to serve as the 5' UTR of mRNA vaccines.
