快速分离柱-高效液相色谱法测定头发中的尼古丁和可天宁

Study on determination of nicotine and cotinine in human hair by HPLC with rapid column

  • 摘要: 研究了用快速分离柱-高效液相色谱测定头发中尼古丁和可天宁的方法.样品中的尼古丁和可天宁用ZORBAX Stable Bound(Φ4.6 mm×50 mm,1.8μm)C18快速分离柱为固定相,15%的乙腈(内含0.02 mol/L十二烷基磺酸钠和0.05 mol/L pH值为3.0的磷酸盐缓冲)为流动相,紫外检测波长为260 nm,流速为2.0mL/min.在该色谱条件下,尼古丁和可天宁在1.0 min内即可达到基线分离.方法标准回收率94%~104%,相对标准偏差分别为2.3%和2.4%.新方法应用于主动吸烟和被动吸烟者头发中尼古丁和可天宁的测定,结果令人满意.

     

    Abstract: A determination of nicotine and cotinine in human hair with HPLC with rapid column was reported.The rapid column was ZORBAX Stable Bound(Φ4.6 mm×50 mm,1.8 μm).The mobile phase contained 15% acetonitrile,0.02 mol/L SDS and 0.05 mol/L pH=3.0 phosphate buffer.The recoveries of nicotine and cotinine were 94%—104% and the relative standard deviations were 2.3% and 2.4%,respectively.
