Analysis of endophytic bacterial diversity in seven pear varieties based on high-throughput sequencing

  • 摘要: 目前对于梨内生细菌多样性的研究几乎为零,这在很大程度上限制了梨内生细菌资源的开发和利用. 为了探明梨内生细菌多样性,挖掘潜在的微生物资源及功能,研究采用高通量测序技术测定7个不同品种梨内生细菌菌群16S rRNA基因V4~V5区序列,并进行生物信息学分析. 在7个不同品种梨中获得有效序列数为30001~42255条,并聚成33~99个可操作分类单元(OTU),涉及17个门、28个纲、52个目、85个科以及120个属. 此外,7个梨品种内生细菌的Shannon指数、Simpson指数和Chao指数分别在0.481~1.154、0.452~0.730和42.333~103.875之间,说明测定的7个不同品种梨含有丰富的内生细菌种类,其中丰水梨和水晶梨内生细菌最为丰富. 不同分类阶元的统计结果表明,不同品种的梨在门、纲、目、科、属水平上的主要内生细菌类型相同,但具体占比不同. 7种梨内生细菌多样性均较为丰富,且不同梨品种菌群结构存在一定差异. 研究为进一步揭示梨与内生细菌之间的互作关系提供参考,为后续研究奠定基础.

     

    Abstract: Pyrus spp. has a long history of planting, which is rich in varieties, high in nutritional and medicinal value. At present, there are few studies on the endophytic bacterial diversity of pear, which limits the development and utilization of pear endophytic bacterial resources. To explore the diversity of endophytic bacteria in pears and discover the potential microbial resources and functions, high-throughput sequencing was used to determine the V4-V5 region of 16S rRNA gene of endophytic bacterial community in seven pear varieties, and bioinformatics analysis was carried out. Among the seven varieties, 30 001−42 255 effective sequences were obtained, and 33−99 operational taxonomic units (OTUs) were clustered. These sequences were divided into 17 families, 28 classes, 52 orders, 85 families and 120 genera. In addition, the Shannon index, Simpson index and Chao index of endophytic bacteria in seven pear varieties were 0.481−1.154, 0.452−0.730 and 42.333−103.875, respectively. The data indicated that the endophytic bacteria of the seven tested varieties were rich, Fengshui pear and Crystal pear had the most abundant endophytic bacteria. In different taxonomic categories, the main endophytic bacteria types of different pear varieties were the same at the level of phylum, class, order, family and genus, but the practical proportion is different. The diversities of endophytic bacteria in the seven pear varieties were ralatively rich, and there were some differences in the bacterial community structure of different pear varieties. The reference for further revealing the interaction between pear and endophytic bacteria was provided.
