Abstract:
An analytical method was established for the determination of 6 kinds of anthocyanins in “Zijuan” tea with high performance liquid chromatography (HPLC). The samples were extracted using a deep eutectic solvent (DES) prepared with choline chloride and oxalic acid (molar ratio 1∶2), after ultrasonic extraction at 60 ℃ for 20 minutes, the content of six anthocyanins in "Zijuan" tea was quantitatively analyzed using an external standard method. The calibration curves of the six anthocyanins showed good linearity with correlation coefficients of above 0.999 4 in the range of 0.1−5 mg/L. Limit of the detection (LOD) and the limit of the quantitative (LOQ) were 0.2−0.5 mg/g and 0.8−1.7 mg/g respectively. The recoveries at 0.5, 2.5, 5.0 mg/kg were 70.4%−94.3% with relative standard deviations of 1.1%−11.3%. The established method of ultrasound assisted deep eutectic solvent extraction and high-performance liquid chromatography for analyzing anthocyanins in “Zijuan” tea has good specificity, is simple and environmentally friendly, and has better extraction efficiency than traditional methods. This research provides a new method for extracting anthocyanins from “Zijuan” tea, which has good development prospects and application potential.
