小豹斑鹅膏遗传多样性研究

A study on the genetic diversity of Amanita parvipantherina

  • 摘要: 小豹斑鹅膏子实体形态多样,不同地域所采标本有较大差异.作者应用ISSR及ITS分子标记,对采自12个不同地区的小豹斑鹅膏居群进行了遗传多样性及聚类分析.结果表明,12条ISSR引物扩增的多态性条带比例(PPB)为72.84%,居群间的基因分化系数(Gst)为0.426 1,表明小豹斑鹅膏具有丰富的基因多样性,其中57.4%存在于居群内,42.6%存在于地理隔离较远的居群间;12个地区样品的IST序列长度皆为710 bp,有30个碱基位点在ITS1区发生变异.2种方法的遗传距离分析及聚类结果基本相似,都表明小豹斑鹅膏的遗传分化与地域的相关性较大.ISSR揭示遗传分化及遗传变异的能力较ITS强.

     

    Abstract: The sporophores of Amanita parvipantherina are highly diversified by different collecting areas.The genetic diversity and clustering analysis of A.parvipantherina populations collected from 12 different areas were carried by using ISSR and ITS molecular markers in this study.The results were as follows:A.parvipantherina had high gene diversity,of which,57.4% existed within populations,and 42.6% among populations with geographical isolation,as the percentage of polymorphic band(PPB) amplified by 12 ISSR primers was 72.84%,and Gst among population was 0.426 1;The length of ITS sequences from the 12 samples was all 710 bp,and 30 base locus of ITS1 region had mutation;The results of both genetic distance analysis and clustering analysis were similar,which indicated the correlation between the genetic differentiation of A.parvipantherina and their geographical distribution;To the ability of revealing the genetic variation and genetic differentiation in this study,ISSR showed stronger resolution than ITS.
