58型HPV E6蛋白的原核表达与分离纯化

  • 摘要: 宫颈癌是一种在女性人群中的常见肿瘤,严重威胁着女性的生理和心理健康;而人类乳头瘤状病毒(HPV)的长期感染是宫颈癌发生的主要诱因.因此,HPV病毒的研究对于我们了解和防治宫颈癌有着十分积极的意义.目前,国外的研究比较多的针对于16型和18型的HPV病毒;但是对于我国比较常见的58型HPV病毒却研究甚少.所以,本试验依据实验室现有的表达纯化平台和噬菌体展示多肽筛选平台,设计并构建了HPV58 E6蛋白的原核表达体系,并对E6重组融合蛋白进行了诱导表达.通过对表达获得的包涵体蛋白进行反复的洗涤、亲和层析以及复性等纯化步骤,得到了纯度较高的E6融合蛋白.

     

    Abstract: Cervical cancer,the second most common cancer in female,is a serious threat to women's health.And the persistent infection of HPV is efficient to cause cervical cancer.Consequently,the research on HPV is important to the fight against cervical cancer.The most current researches on HPV are focused on the HPV16/18,however,the HPV58,the third prevalent type of HPV in China,is ignored by the most researcher abroad.Depending on The existing experimental platform,we designed and constructed the prokaryotic expression plasmid,pET32a-HPV58E6,induced the expression of Trx-E6 fusion protein in BL21(DE3),and purified the fusion protein.
